| I. Background | |
TIGIT 又稱 WUCAM、Vstm3、VSIG9，是一種共抑制受體，屬于免疫球蛋白超家族。 TIGIT 在效應(yīng) CD4+和 CD8+T 細(xì)胞、調(diào)節(jié)性 T 細(xì)胞和 NK 細(xì)胞上可觀察到高水平的表 達(dá).TIGIT 的配體包括 CD112 和脊髓灰質(zhì)炎病毒受體（PVR，也被稱為 CD155、Necl-5 和 Tage4），其中 PVR 是 TIGIT 的高親和同源受體。此外，TIGIT 與 CD226 (DNAM-1)和 CD96 (TACTILE)競(jìng)爭(zhēng)配體。TIGIT 可以有效阻斷 CD155 與 CD96 或 CD226 的結(jié)合，進(jìn)一 步證明了 TIGIT 對(duì) CD155 具有最高的親和力。TIGIT 不僅與 CD226 競(jìng)爭(zhēng)配體，還可以直接 順式結(jié)合 CD226 阻止其同二聚化，使 CD226 無(wú)法與 CD155 結(jié)合發(fā)揮共刺激作用。 | |

TIGIT&PVRIG Dual Effector Reporter Cell 報(bào) 告 基 因 藥 靶 模 型 很 好 的 模 擬 了 體 內(nèi) TIGIT&PVRIG 的信號(hào)轉(zhuǎn)導(dǎo)過(guò)程，原理見(jiàn)下圖所示。
